Targeted Agents for the Treatment of Melanoma: An Overview

چکیده
Malignant melanoma is an important healthcare issue. The incidence of melanoma has in‐ creased dramatically during the last few decades and melanoma is now one of the most com‐ mon forms of cancer. This aggressive form of skin cancer is characterized by its high capacity for invasion and metastasis. Patients with advanced stages of the disease have universally poor prognoses with a median survival of 3–11 months, depending on various prognostic factors. Primary cutaneous melanomas are divided into four groups on the basis of histopathology: su‐ perficial spreading melanoma, nodular melanoma, lentigo maligna melanoma, and acral lenti‐ ginous melanoma. Superficial spreading melanoma is the most common form. In the early growth phase, tumor cells spread laterally into the epidermis, forming an irregular pattern with an uneven color on the skin. Nodular melanoma occurs most often on the chest or back where it has a tendency to grow vertically into the skin, penetrating deeply if not removed. Nodular melanoma lesions are often characterized by a darkly pigmented lump on the skin surface. Len‐ tigo maligna melanoma most commonly occurs on the facial areas of elderly people. It grows slowly and may take several years to develop. Acral lentiginous melanoma is usually found on the palms of the hands, the soles of the feet, or around the toenails. Melanoma is usually curable in its early stages; however, the disease may be fatal if it spreads to other parts of the body. Ma‐ lignant melanoma usually develops via the transformation and proliferation of melanocytes that reside in the basal cell layer of the epidermis. Melanoma can also develop when melano‐ cytes no longer respond to normal cellular growth control mechanisms, which allows the tumor cells to metastasize into nearby tissues or other organs, where they invade and compromise or‐ gan function. Thus, the development of alternative treatment for melanoma is critically impor‐ tant. Recent advances in oncology have led to novel therapeutics that is capable of targeting

An investigation into the photothermal effects of multi- functional gold coated Fe3O4 Nanoparticles in the presence of external magnetic field and NIR laser irradiation on model of melanoma cancer cell line B16F10 in C57BL/6 mice
Introduction: Photothermal therapy using gold nanoshells is one of cancer therapy methods. Gold nanoshells generally consist of a silica core and a thin gold shell. Fe3O4@Au core-shell can be used for magnetic targeted therapy. The objective of this study was investigation of the photothermal effects of magnetically targeted Fe3O4@Au NPs and NIR laser irradiation on model of me...
متن کاملSystemic Targeted Alpha Radiotherapy for Cancer
Background: The fundamental principles of internal targeted alpha therapy for cancer were established many decades ago.The high linear energy transfer (LET) of alpha radiation to the targeted cancer cellscauses double strand breaks in DNA. At the same time, the short range radiation spares adjacent normal tissues. This targeted approach complements conventional external beam radiotherapy and ch...
متن کاملIn vivo Modeling and Molecular Characterization: A Path Toward Targeted Therapy of Melanoma Brain Metastasis
Brain metastasis (B-Met) from melanoma remains mostly incurable and the main cause of death from the disease. Early stage clinical trials and case studies show some promise for targeted therapies in the treatment of melanoma B-Met. However, the progression-free survival for currently available therapies, although significantly improved, is still very short. The development of new potent agents ...
متن کاملAn overview of nanoparticles and their application to drug delivery in cancer: review article
Today, nanoscience has grown and developed in various medical and therapeutic areas, including cancer treatment. On the other hand, cancer and its types have been rumored and inclusive and many people suffer from this fatal and deadly disease. Currently, existing therapeutic method, including chemotherapy, radiotherapy, and etc., along with the therapeutic effect, cause complications that are u...
متن کاملAdvanced Malignant Melanoma: Immunologic and Multimodal Therapeutic Strategies
Immunologic treatment strategies are established in malignant melanoma treatment, mainly focusing on Interleukin-2 in advanced disease and interferon alpha in the adjuvant situation. In advanced disease, therapies with IL-2, interferon and different chemotherapeutic agents were not associated with better patient survival in the vast majority of patients. Therefore, an overview of novel immunolo...
متن کاملImmunotherapy of Melanoma.
Arising from melanocytes in skin, mucosal membranes, eye, and meninges, melanoma is a tumor that has been associated with poor prognosis in advanced disease stages. Given the poor response to chemotherapy and radiation therapy, new treatment approaches with targeted therapy, immunotherapy, and adoptive T-cell therapy have revolutionized the standard of care for patients with advanced melanoma. ...
